目的:提供复方天山堇菜颗粒研究用的化学对照品。方法:分别对干燥的天山堇菜和睡莲化用70%乙醇提取,提取浓缩液得2个提取物Ⅰ a和Ⅱ b,然后分别用水混悬,先后用乙酸乙酯和正丁醇萃取,浓缩正丁醇液部分,得正丁醇提取物Ⅰ b和Ⅱ b,Ⅰb再经聚酰胺层析.乙醇-水(4:6)洗脱得七叶内酯,Ⅱb再经硅胶和C18柱层析,得到烟花苷。结果:2个化合物的UV、IR、1H—NMR和13C—NMR数据符合文献值。结论:提取得到的七叶内酯和烟花苷作为化学对照品是可行的。
Abstract:Objective: The method was established in extract two chemical reference substances from the whole plants of Viola tianshanica Maxin. and the flowers of Nymphaea Candida presl. respectively. Method: The dried whole plants of Viola tianshanica Maxin. ( Ⅰ ) and the flowers of Nymphaea Candida presl.. ( Ⅱ ) were extracted with 70% ethanol, respectively. The 70% aq. EtOH extracts were condensed in vacuum to give two crude extracts ( Ⅰ a and Ⅱ a). The extracts of Ⅰa, Ⅱ a were suspended in water and portioned with petroleum ether (or EtOAC) and n-BuOH. The n-BuOH fraction were evaporated under reduced pressure to obtain extracts of Ⅰ b and Ⅱ b. The Ⅰ b extraction was subjected to polyamide column chromatography, elutcd with EtOH-H2O(4 : 6) to give chemical reference substance of esculetin. The Ⅱ b extraction was subjected to silica gel and C18column chromatography to obtain second reference substance of nicotiflorin. Results: The spectroscopic data (UV, IR, H-NMR and 13C-NMR) of two reference substances were agreed with reports of literatures. Conclusion: The structure of two compounds is corrective. It is suitable for chemical reference substance.
