目的探讨复方一枝蒿微丸的抗炎、免疫及体内外抗菌作用。方法采用小鼠耳廓肿胀和毛细血管通透性试验观察复方一枝蒿微丸的抗炎作用;检测其对SRBC免疫小鼠血清中IgM生成的影响,以及对LPS及Con A诱导下T、B淋巴细胞增殖作用的影响;测定其对金黄色葡萄球菌、肺炎链球菌的体外抗菌作用,并观察其对金黄色葡萄球菌、大肠杆菌感染小鼠死亡的保护作用。结果复方一枝蒿微丸低、中、高(0.5、1.0、2.0 g/kg)剂量对二甲苯所致小鼠耳廓肿胀和腹腔毛细血管通透性升高均有显著的抑制作用;可显著升高SRBC免疫小鼠血清IgM水平;在232~15 000μg/m L浓度范围内,能显著抑制Con A诱导的T淋巴细胞增殖作用(P<0.05);在464~15000μg/m L浓度范围内,能显著抑制LPS诱导的B淋巴细胞增殖作用(P<0.05);1.0 g/m L的复方一枝蒿微丸对金黄色葡萄球菌、肺炎链球菌抑菌圈直径分别为1.65、1.38 mm,MIC分别为62.5、6.4 mg/m L,MBC分别为125.0、12.5 mg/m L,且能显著降低金黄色葡萄球菌、大肠杆菌感染小鼠的死亡率(P<0.05)。结论复方一枝蒿微丸具有一定的抗炎、抗菌和调节免疫功能的作用。
Abstract:Objective To investigate the anti-inflammatory,immune and antibacterial effects in vitro and in vivo of the compound Yizhihao pellets. Methods The anti-inflammatory effect of compound Yizhihao pellets was observed through the ear edema and capillary permeability in mice. Impact of generated of IgM of SRBC immune mice in the serum and proliferation effect of LPS and Con A induced T,B cell was measured. The antibacterial effect of compound Yizhihao pellets on Staphylococcus aureus,streptococcus pneumonia in vitro was detected. Evaluation of the immune effect in mice against Staphylococcus aureus,Escherichia coli challenge. Results Compound Yizhihao pellets dosage( 0. 5,1,2 g / kg) could significantly inhibit ear edema and capillary permeability increase in mice model and increase IgM level of SRBC immunized mice in the serum. It could significantly inhibit the proliferation of T cells induced by Con A( P <0. 05) at 232 ~ 15 000 μg / m L and inhibit the proliferation of B cells induced by LPS( P <0. 05) at 464 ~ 15 000 μg / m L. When concentration of Compound Yizhihao pellets was 1. 0 g / m L,diameter of bacteriostatic ring of Staphylococcus bacteria and Streptococcus pneumoniae were 1. 65 mm and 1. 38 mm,MIC were 62. 5,6. 4 mg / m L,MBC were 125. 0,12. 5 mg / m L,and it could significantly reduce the mortality of mice infected with Staphylococcus aureus and Escherichia coli( P <0. 05). Conclusion Compound Yizhihao pellets has certain effects of anti-inflammatory,antibacterial and regulating immune function.
