目的比较维吾尔族和汉族散发性乳腺癌患者乳腺癌1号基因(BRCA1)启动子区域甲基化水平及其与临床病理特征的关系。方法采用甲基化特异性PCR法检测维吾尔族和汉族乳腺癌患者BRCA1基因启动子区域甲基化状态,分析甲基化状态改变与乳腺癌发生、发展的关系。结果汉族患者甲基化率为26%,维吾尔族患者甲基化率为30%,汉族与维吾尔族患者甲基化率差异无统计学意义(P=0.65)。BRCA1基因甲基化组与非甲基化组临床病理特征的比较中,汉族患者的年龄及淋巴结状态与BRCA1甲基化有关(P分别为0.021及0.001),维吾尔族患者仅组织学分级与BRCA1甲基化有关(P=0.001)。结论 BRCA1基因表观遗传学改变在肿瘤的发生、发展过程起着重要作用,且与散发性乳腺癌患者的发病年龄及组织学分级和淋巴结状态的发生密切相关。
Abstract:Objective To compare the methylation level of breast cancer gene(BRCA1)promoter region and its relationship to clinical pathological features in the patients with sporadic breast cancer of Uygur and Han patients.Methods The methylation status of BRCA1 gene promoter was detected by methylation specific PCR technique,and the relationship between methylation status and the development of breast cancer was analyzed.Results The methylation rate was 26%in Han patients while it was 30%in Uygur patients,that there was no significant(P =0.65).The age and lymph node status of the patients with BRCA1 methylation and BRCA1 methylation in the BRCA1 gene methylation group were compared with the clinical pathological features(P =0.021 and 0.001).Conclusion Epigenetic changes of BRCA1 gene plays an important role in the development of tumor,and is closely related to the age of onset of the disease and the incidence of lymph node status in patients with sporadic breast cancer.
