基于~1H-NMR研究罗勒总黄酮对ApoE-/-小鼠血清代谢组学的影响

摘要:

目的采用核磁共振氢谱(1 H-NMR)研究罗勒总黄酮对ApoE-/-小鼠血清代谢组学的影响。方法选择6w龄雄性ApoE-/-小鼠20只随机分为2组:模型组(蒸馏水10mL/kg体质量)、罗勒总黄酮组(罗勒总黄酮240mg/kg体质量),高脂饲料喂养;将6 w龄相同遗传背景的雄性C57BL/6J小鼠10只作为空白组(蒸馏水10mL/kg体质量),普通饲料喂养。灌胃给药14w后取各组小鼠血清,采用1 H-NMR检测小鼠血清中的主要代谢物,再利用OPLS-DA和PLS-DA方法对1 H-NMR图谱进行分析,筛选小鼠血清主要差异代谢物。结果与空白组相比较,模型组小鼠血清中脂类、VLDL、LDL、脯氨酸、β-羟丁酸、甲胺、肌醇、β-葡萄糖、α-葡萄糖的含量增多,差异有统计学意义(P<0.05);而亮氨酸、异亮氨酸、缬氨酸、丙氨酸、谷氨酰胺、甘氨酸、赖氨酸、牛磺酸、乳酸、肉碱、胆碱、柠檬酸、丙酮酸、肌酸的浓度水平有不同程度的下降,差异有统计学意义(P<0.05)。与模型组相比较,罗勒总黄酮组对小鼠血清上述代谢物均有不同程度逆转作用。结论罗勒总黄酮可能通过调节ApoE-/-小鼠的能量、氨基酸、脂质代谢途径,对动脉粥样硬化体内代谢紊乱起到一定调控作用。

Abstract:

Objective To observe the effects of total flavonoids of Ocimum Basilicum L.on serum metabolites in apolipomtein E knockout(ApoE-/-)mice by using 1 H-NMR-based metabonomics.MethodsTwenty six-week-old male ApoE-/-mice were divided into two groups:model group(distilled water,10 mL/kg,i.g)and OBL group(Ocimum Basilicum L.flavonoids,240 mg/kg,i.g),fed with high fat diet.The homogeneous male mice of C57 BL/6 Jwere used as the normal group(distilled water,10 mL/kg,i.g)and fed with normal diet.After 14 weeks,the variability of serum metabolites were analyzed by 1 HNMR-based metabonomics and compared by OPLS-DA and PLS-DA.Results Compared with the normal group,the model group showed a significant increase in the serum levels of Lipid,VLDL,LDL,Proline,β-Hydroxyisobutyrate,Methylamine,Myoinositol,β-glucose andα-glucose,and showed a significant decrease in the serum levels of Leucine,Isoleucine,Valine,Alanine,Glutamine,Glycine,Lysine,Taurine,Lactate,Carnitine,Choline,Citrate,Pyruvate and Creatine.Compared with the model group,serum metabolites levels of the above were changed inversely in the OBL group(P <0.05).Conclusion Total flavonoids of Ocimum Basilicum L.may have anti-atherosclerotic effects by regulating the glucose,lipid and amino acid metabolism in ApoE-/-mice.
