目的探究发酵中药复方水剂化糖酶对糖尿病小鼠血糖的影响。方法取雄性昆明种小鼠,体质量(22.0±2.0)g,腹腔注射小剂量链脲佐菌素(STZ)建立糖尿病小鼠模型,1w后根据空腹血糖结果随机分为模型组、二甲双胍组、化糖酶低、高剂量组并予以相应药物,每天1次,5w后测定小鼠体质量、空腹血糖(FBG)、血清胰岛素、糖耐量(OGTT)。结果 STZ注射后1w,小鼠血糖值变化明显且稳定,综合体质量变化及饮水、尿量等情况,模型成功纳入64只。与模型组小鼠比较,化糖酶低、高剂量组小鼠体质量减轻;药物干预4w后,与模型组小鼠比较,化糖酶低、高剂量组小鼠饮水量减少,差异有统计学意义(P<0.05);第3w开始,与模型组小鼠比较,化糖酶低、高剂量组小鼠空腹血糖降低,差异有统计学意义(P<0.05);与模型组小鼠比较,化糖酶高剂量组小鼠血清胰岛素水平降低,差异有统计学意义(P<0.05);与模型组小鼠比较,化糖酶低、高剂量组小鼠口服葡萄糖后4个时间点的血糖降低,差异有统计学意义(P<0.05)。结论发酵中药复方水剂化糖酶对STZ所致糖尿病小鼠的血糖有较好的调节作用。
Abstract:Objective To explore the effects of fermented Chinese herbal compound Huatangmei(HTM)on blood glucose in diabetic mice.Methods A diabetic mouse model was established by intraperitoneal injection of small dose of streptozotocin(STZ)in male KM mice weighing within(22±2)g.One week later,the treatment mice were randomly divided into model group,metformin group,low-and high-dose HTM group with corresponding medicine orally once a day,after 5 weeks the changes of mice in body weight,fasting plasma glucose(FBG)and serum insulin were measured.Results After one week of STZ injection,the blood sugar value was obvious increased and tended to be stable,and combined with the weight changes and water-drinking,urine volume changing as well,the model was successfully included by64 mice.Compared with the model group,low-and high-dose HTM improved the weight loss of mice with a little trend;after 4 weeks of intervention,the amount of water mice drunk in the low-and high-dose HTM groups were reduced significantly(P <0.05);starting from the third week,the FBG in low-and high-dose HTM groups were decreased(P <0.05);high dosage of HTM could obviously suppress the serum insulin level(P <0.05);and the result of glucose tolerance OGTT also showed low-and high-dose HTM could reduce the blood glucose at 4 time-points after oral administration(all P <0.05).ConclusionFermented Chinese herbal compound Huatangmei can regulate the blood glucose of diabetic mice well.
