摘要:

目的探讨两色金鸡菊黄酮成分马里苷对胰岛β细胞的保护作用及可能机制。方法将培养的MIN6细胞分为正常对照组、高糖高脂模型组及马里苷低、中、高浓度(100、200、400μmol/L)给药组。运用四甲基偶氮唑盐比色法(MTT)检测其活性;酶联免疫法(ELISA)检测胰岛素分泌量;Western Blot方法检测细胞中BAX,Bcl-2蛋白表达水平。结果与正常对照组比较,高糖高脂模型组细胞增殖能力能力明显降低(P<0.05),马里苷不同剂量干预组MIN6细胞增殖均明显增加(P<0.05)。与正常对照组比较,高糖高脂模型组细胞胰岛素分泌量明显降低(P<0.05);与高糖高脂模型组比较,给药组细胞胰岛素分泌量明显升高(P<0.05)。Western印迹显示,与正常对照组比较,高糖高脂模型组BAX蛋白表达量增多,给药组随浓度递增依次减少;高糖高脂模型组Bcl-2蛋白表达量减少,给药组随浓度增加依次增多(P<0.05)。结论马里苷对高糖高脂造成的胰岛细胞损伤有保护作用,其可能机制可能与Bcl蛋白家族有关。

Abstract:

Objective This study aimed to investigate the protective effects of Coreopsis basalis flavonoids MAREIN on pancreatic beta cells and its possible mechanism.Methods The cultured MIN6 cells were divided into normal control group,high glucose and high fat model group,high sugar and high fat with different concentration of the intervention groups.Four methyl azo salt colorimetric method(MTT)was used to detect cell activity;ELISA method was used to detect the insulin secretion;Blot Western method was used to detect the expression level of BAX and Bcl-2in the cells.Results Compared with the control group,the cell survival rate of the model group was significantly lower(P <0.05),and the proliferation of MIN6 cells in the intervention group were significantly increased(Pall<0.05).Compared with the normalgroup,the insulin secretion in the model group was significantly decreased(P <0.05),and the insulin secretion was significantly increased in the group with high glucose and high fat group(P <0.05).Western blot showed that,compared with the control group,the expression of BAX protein increased in model group,but decreased with the increasing of the concentration of the drug in the drug groups;Bcl-2protein decreased in model group,but increased with the increase of concentration of the drug in the drug groups(P <0.05).Conclusion The protective effect of the on the injury of islet cells induced by high glucose and high fat may be related to the Bcl protein family.
