ANXA6基因多态性与门冬氨酸氨基转移酶水平的相关性研究

摘要:

目的 探讨ANXA6基因SNP1(rs2303022)位点与门冬氨酸氨基转移酶(AST)水平的相关性。方法 收集2018年10-11月在新疆医科大学健康体检中心体检的7 095名学生为研究对象,采用极端表型研究策略,对测定的不同性别人群的低密度脂蛋白胆固醇水平进行从小到大的排序,根据纳入排除标准筛选后最终纳入792例,测定血尿素氮(BUN)、肌酐(Cr)、谷丙转氨酶(ALT)、门冬氨酸氨基转移酶(AST)、总胆红素(TBIL)、空腹血糖(FBG)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C),提取血液样本DNA,采用SNPscan技术进行基因型分型,对收集数据进行统计学分析。结果 病例组和对照组性别分布、BMI、ALT、Cr、FBG、TC、LDL-C、HDL-C、TG水平比较,差异有统计学意义(P<0.05)。男性对照组和男性病例组在BMI、ALT、TC、LDL-C、TG水平上的差异有统计学意义(P<0.05);女性对照组和女性病例组在年龄和ALT水平上的差异有统计学意义(P<0.05)。男性对照组和男性病例组ANXA6基因rs2303022在隐性模型(CC+CG;GG)中的分布频率差异有统计学意义(P<0.05)。Logistic回归分析结果显示,AST与ALT的水平呈正相关(OR=1.18,P<0.001,95%CI:1.14~1.22),年龄和HDL-C水平是AST升高的影响因素。结论 本研究未发现ANXA6基因SNP1(rs2303022)位点与AST相关,AST升高可能与年龄和HDL-C水平变化有关。

Abstract:

Objective Discuss the correlation between SNP1(rs2303022) site of ANXA6 gene and the level of aspartate aminotransferase(AST). Methods 7 095 students who were examined in the Health Examination Center of Xinjiang Medical University from October to November 2018 were collected as research objects. The extreme phenotype research strategy was used to rank the LDL cholesterol levels measured by different gender groups from small to large. 792 cases were finally included after screening according to the inclusion and exclusion criteria, and blood urea nitrogen(BUN), creatinine(Cr), alanine transaminase(ALT), aspartate aminotransferase(AST), total bilirubin(TBIL), fasting blood glucose(FBG), triglyceride(TG), total cholesterol(TC), low-density lipoprotein cholesterol(LDL-C) and high-density lipoprotein cholesterol(HDL-C), and the DNA was extracted from the blood samples. SNPscan technology was used for genotyping, and the collected data was statistically analyzed. Results There were a statistically significant difference in gender distribution, BMI, ALT, Cr, FBG, TC, LDL-C, HDL-C, and TG levels between case and control groups(P<0.05). There was a statistically significant difference in BMI, ALT, TC, LDL-C, and TG levels between male control group and male case group(P<0.05). There were statistically significant differences in age and ALT levels between female control group and female case group(P<0.05). There was a statistically significant difference in the distribution frequency of the ANXA6 gene rs2303022 genotype between male control group and male case group in the recessive model(CC+CG; GG)(P<0.05). The results of logistic regression analysis showed a positive correlation between AST and ALT levels(OR=1.18, P<0.001, 95%CI: 1.14-1.22), age and HDL-C levels were the contributing factors for the increased AST. Conclusion The SNP1(rs2303022) site in the ANXA6 gene was not associated with AST, and the increased AST may be related to changes in age and HDL-C levels.
