目的:构建Lewis肺癌C57BL/6小鼠皮下移植瘤模型,研究恩度与顺铂及联和用药对Lewis肺癌C57BL/6小鼠皮下移植瘤的生长抑制作用。方法:使用C57BL/6小鼠作为实验动物,培养小鼠Lewis肺癌细胞,收集对数生长期的肿瘤细胞,调整细胞悬液浓度至1×107/ml。于小鼠右侧腋部皮下注射细胞悬液0.2 ml(细胞总数为2×10⁶),建立Lewis肺癌小鼠皮下移植瘤动物模型。待接种后第8天,能扣及肿瘤时(约8 mm)肝始实验。将40只荷瘤小鼠随机分为四组。A组:生理盐水组;B组:顺铂组;C组:恩度组;D组:恩度与顺铂联用组。C组、D组每日给予恩度3 mg/kg,腹腔注射给药,每日1次,连续14 d;B组、D组腹腔注射顺铂0.2 m1(按照1 mg/kg给药),每天1次,连续2周。A组给予生理盐水0.2 ml/只腹腔注射,每日1次,连续二周后处死。观察各组小鼠的生存质量情况,每3天称取体重一次并测量肿瘤长短径,描绘肿瘤生长曲线;称量瘤重,计算抑瘤率。结果:各治疗组小鼠移植瘤生长较生理盐水组相对缓慢,联合治疗组肿瘤体积明显小于生理盐水组、顺铂组和恩度组;顺铂组、恩度组和联合治疗组抑瘤率分别为32.19%、21.96%、55.08%。顺铂与恩度联用对小鼠移植瘤具有协同抑制作用(Q>1.15)。结论:在实验条件下,恩度与化疗药物顺铂均对Lewis肺癌皮下移植瘤生长有明显抑制作用。恩度与顺铂对Lewis肺癌移植瘤具有较好的协同抑制作用,同时能够改善小鼠的生存质量,在肺癌治疗中具有一定潜力。

Objective:To stody the function of YH-16,DDP and combined Using on subutaneouslv implanted Lewis lung carcinoma(LLC) of C57BL/6 Mice.Methods:Forty C57BL/6 miee weighting 20±2,6～8 weeks old were used and all were male.The LLC cell line was cultivated in a humidified atmosphere of 5 CO₂ at 37℃in DMEM medium,supplemented with 15%calf serum and antibiotics.Tumor models were established by hypodermical injection of 2×10⁶ tumor cells into the right armpit of the mice,and tumor growth was calculated by measuring the long and short diameters.The mice were divided randomly into four groups and eaehgroup consists of 10 miee.All groups were treated from the eighth day of the LLC cells injected throughout the whole experiment.Cisplatin group were given with DDP 0.2 ml(1 mg/kg), YH 16 group were given with YH-16 1.5 mg/kg,DDP+YH-16 group were given with DDP+YH-16 the same dose like DDP group and YH-16 group,and equal volume of saline was given to control group meantime. Tumors growth was delineated the tumor growth curve.Three weeks after implantation,all mice were sacrificed.The weight of tumors and mice was measured.Results.The transplant lump volume of treated groups were obvious small than the saline group.DDPgroup,YH 16 group,DDP+YH-16 group. Their suppressing rate are 32.19%,21.96%,55.08%.Conclusion.Under the experiment condition,DDP and YH-16 can repress implanted lung carcinoma obviously,they have conjunction function(Q>1.15). They can improvement living quality of small rats.
